Abstract
Lymphomas are a diverse group of cancers that originate from lymphoid tissues, encompassing a broad spectrum of subtypes with distinct molecular, histological, and clinical features. This chapter discusses the epidemiological trends, risk factors, and molecular mechanisms underlying lymphoma development. In 2023, non-Hodgkin lymphoma was the seventh most frequently diagnosed cancer among men and sixth among women in the United States with a 4% incidence in both sexes, making it the ninth leading cause of cancer-related deaths, contributing to 3%-4% of all diagnoses. Hodgkin lymphoma represents approximately 0.4% of new cancer cases worldwide with incidence peaks among young adults and older individuals. Advancements in diagnostic techniques, including histopathology, immunohistochemistry, flow cytometry, and next-generation sequencing, have significantly improved the accuracy of lymphoma diagnosis and subclassification. The role of key molecular markers and relevance of biomarker-driven approaches in disease stratification are also discussed. Therapeutic innovations have transformed lymphoma management with a shift from conventional chemotherapeutic regimens toward targeted therapies, monoclonal antibodies, immune checkpoint inhibitors, and chimeric antigen receptor T-CELL therapies. The emergence of precision medicine driven by genomic profiling and personalized treatment approaches is revolutionizing patient care and improving prognosis. Furthermore, the chapter highlights the challenges of treatment resistance, relapse, and the need for novel therapeutic agents to overcome these barriers.
